it is not a topic of the number of NICs - you should take a look at
their configuration.
Regarding your config:

This route command makes currently no sense to me (or you didn't post
all information). Maybe it interferes with the config of you NICs
(espacially the 2nd one) -- but without knowing its detailed config I
couldn't tell you.
